It may have both an anti-spirochaete activity and an anti-inflammatory activity, similar to the treatment of rheumatoid arthritis. And caution is required if patients have certain heart conditions, diabetes, psoriasis etc.

Mechanism of action and resistance, as well as status of resistance. studies indicated that chloroquine resistance was CHLOROQUINE Chloroquine was first synthesized in Germany, but it was not recognized as a potent antimalarial drug until the 1940s during the US World War II military effort. By 1946, Chloroquine, an antimalarial drug, can also be used in the manipulation of the immune system. • This review demonstrates the multitude of actions displayed by CQ, dependent on lysosome dysfunction and also pH-independent mechanisms. • Chloroquine administration ameliorates rheumatoid arthritis, systemic lupus and viral infections. Chloroquine and hydroxychloroquine are drugs derived from the quinoline molecule. Both are used as antimalarial blood schizonticides, and hydroxychloroquine is also frequently used as an antirheumatic. Their mechanism of action is not entirely understood.

The most serious adverse effects affect the eye, with dose-related retinopathy as a concern even after hydroxychloroquine use is discontinued. The most common adverse effects are a mild nausea and occasional stomach cramps with mild diarrhea.

The drug chloroquine is bactericidal for Bacillus megaterium ; it inhibits DNA and RNA biosynthesis and produces rapid degradation of ribosomes and dissimilation of ribosomal RNA. Inhibition of protein synthesis is also observed, evidently as a secondary effect. Inhibition of DNA replication is proposed as a general mechanism of the antimicrobial action of chloroquine. It is a hydroxylated version of chloroquine, with a similar mechanism of action. However, following an identical dose of hydroxychloroquine and chloroquine, tissue levels of chloroquine are 2.5 times those of hydroxychloroquine. Hydroxychloroquine is preferred due to its safer profile. Chloroquine is thought to exert its antimalarial effect by preventing the polymerization of toxic heme released during proteolysis of hemoglobin in the Plasmodium digestive vacuole. The mechanism of this blockade has not been established. We incubated cultured parasites with subinhibitory doses of 3Hchloroquine and 3H quinidine. These 3Hquinoline compounds became associated with hemozoin.
